A technique of separation and identification of components in a mixture used in biochemistry as well as in analytical chemistry which have wide range of applications. In chromatography, the separation occurs between two phases that is mobile and stationary phase. Analyte is the substance which is to separated and a solvent carries it. In HPLC, the sample is forced through a column by a liquid at high pressure. The main principle of separation is adsorption and partition chromatography. The technique is used for determining quantitative and qualitative analysis. A HLPC instrument contains a degasser, sampler, pumps and detector along with digital processing unit to provide data analysis. In pharmaceutical industry, this technique helps in estimating purity.
