Several review articles have shown that deficient DNA repair, allowing greater accumulation of DNA damage, causes premature aging; and that increased DNA repair facilitates greater longevity. healthy aging and longevity in humans are modulated by a lucky combination of genetic and non-genetic factors. family studies demonstrated that about 25 % of the variation in human longevity is due to genetic factors. in humans and other mammals, DNA damage occurs frequently and DNA repair processes have evolved to compensate. if DNA damage is the underlying cause of aging, it would be expected that humans with inherited defects in the ability to repair DNA damages should age at a faster pace than persons without such a defect. numerous examples of rare inherited conditions with DNA repair defects are known.
